Thabo started playing for the junior Swiss national basketball team and began a professional career with Vevey Riviera Basket at the age of 17. After that, he played for Elan Chalon in France and spent his first year with the junior under-21 squad. In 2002, he earned a rotation spot on the senior club and played 30 games.
During this period, he averaged 4 points, 3.5 rebounds, and 1 assist per game. After that, he earned a starting spot on his team and led them to a third-place finish in the French league. He signed with the Italian team Angelico Biella after a contract dispute between him and the Elan Chalon team. In the 2006 NBA draft, the Philadelphia 76ers picked him but traded him to the Chicago Bulls for Rodney Carney.

The club then traded him to the Oklahoma City Thunder on February 19, 2009. Moreover, he was selected to the All-NBA Defensive Second Team for the 2009-20 NBA season. Turkish team Fenerbahce Ulker signed him for Euroleague matches during the 2011 NBA lockout in October 2011.

After that, he signed a one-year deal with the Houston Rockets on September 23, 2019. On March 31, 2021, he announced his retirement from professional basketball. Sefolosha was arrested with Pero Antic outside a nightclub in New York City on April 8, 2015. The police arrested them for interfering with police after Indiana Pacers player Chris was stabbed in the abdomen in an argument.
During this altercation, the police broke his right leg and he missed the rest of the season. The Manhattan jury found him not guilty of the charges on October 9 and he announced to sue New York City and eight police officers for up to $50 million in damages on October 21.
Later, he settled with the NYPD for $4 million on April 9, 2017, and he donated a substantial portion to Gideon’s Promise. Utah Jazz suspended him without pay for five games for violating Anti-Drug Program on April 3, 2018.
